P257 Microbiological characterisation of methicillin-resistant Staphylococcus aureus isolates recovered from cystic fibrosis people during two Spanish multicentre studies (2013–2021)

To characterize methicillin-resistant S. aureus (MRSA) isolates recovered from cystic fibrosis (CF) people during two Spanish multicenter studies (2013–2021) by whole genome sequencing (WGS). 51 MRSA were prospectively CF (51% female, median age 20 y [26–15]) 17 hospitals 2 (30 in 2013 and 21 2021). Identification was performed MALDI-TOF. WGS (Illumina-Novaseq-6000) carried out. SPAdes Prokka software used for de novo assembly annotation. Genome QUAST bacterial identification confirmed using Kraken. In silico MLST, spa-Typing SCCmec assignment also performed. Resistance genes identified Abricate with ARG-ANNOT Resfinder databases (95% identity/85% coverage). prevalence 11% 9% 2021. Hospital-acquired (HA) clones more prevalent than community-acquired (CA) ones both 2021 (67% vs 33% 71% 29%, respectively) appeared older (median [28–16] Vs. 18 [25–14]). HA-MRSA population dominated CC5, being ST125-MRSA-IVc t067 the most clone (37% of isolates). CA-MRSA showed a higher clonal diversity, ST1-MRSA-IVa t127 frequent clone. Interestingly, PVL-positive belonging to ST8-MRSA-IV detected among CA-MRSA, one each study period. Also, we swine-associated ST398-MRSA-V t011 2013. Regarding resistome, 43 resistance against several families antibiotics detected, including transmissible cfr gene, which confers linezolid.
